1、**内膜上皮细胞菌种传代保存法：

有些微生物当遇到冷冻或干燥等处理时，会很快死亡，因此在这种情况下，只能求助于传代培养保存法。传代培养就是要定期地进行菌种转接、培养后再保存，它是*基本的微生物保存法，例如酸奶等常用生产菌种的保存。传代保存时，培养基的浓度不宜过高，营养成分不宜过于丰富，尤其是碳水化合物的浓度应在可能的范围内尽量降低。培养温度通常以稍低于*适生长温度为好。若为产酸菌种，则应在培养基中添加少量碳酸钙。

一般地，大多数菌种的保藏温度以5℃为好，像***、霍乱弧菌及部分病原**等微生物菌种则可以使用37℃进行保存，而蕈类等大型食用菌的菌种则可以室温直接保存。传代培养保存法虽然简便，但其缺点也很明显，如：①菌种管棉塞经常容易发霉;②菌株的遗传性状容易发生变异;③反复传代时，菌株的病原性、形成生理活性物质的能力以及形成孢子的能力等均有降低;④需要定期转种，工作量大;⑤杂菌的污染机会较多。

2、**内膜上皮细胞菌种液体石蜡覆盖保存法：

该法较前一种方法保存菌种的时间更长，适用于霉菌、酵母菌、放线菌及需氧**等的保存。此法可防止干燥，并通过限制氧的供给而达到削弱微生物代谢作用的目的。其具有方法简便的优点，同时也适用于不宜冷冻干燥的微生物(如产孢能力低的丝状菌)的保存，而某些**如固氮菌、乳酸杆菌、明串珠菌、分枝杆菌、红螺菌及沙门氏菌等和一些**如卷霉菌、小克银汉霉、毛霉、根霉等不宜采用此法进行保存。
