Description: The monoclonal antibody SJBCH recognizes human Skp-2 (S-phase kinase-associated protein 2), a 47 kDa F-box protein that interacts with the Cyclin E-Cdk2 complex. Members of the F-box protein family are characterized by a 40 amino acid F-box motif, named after cyclin F. F-box proteins act as adapter proteins, associating with substate proteins and bringing them to the core of the ubiquitin ligase complex where they bind to Skp-2. Skp-2 associates with p27 and is required for the ubiquitination and subsequent degradation of p27 (cyclin-dependent kinase (Cdk) inhibitor). P27 binds to and inhibits cyclin E-cdk2 complexes, as a result preventing progression through the cell cycle. It is the Skp-2-dependent degradation of phorphorylated p27 that allows for the G1-S transition in both transformed cells and diploid fibroblasts.
Skp-2 has been reported to be overexpressed in a variety of cancer types and correlates with poor prognosis. Skp-2 overexpression has been documented in colon mucinous carcinoma, gastric adenocarcinoma, thymoma, melanoma, prostate adenocarcinoma, lung adenocarcinoma, and ER (estrogen receptor) negative breast cancers.
