ASSAY PRINCIPLE
In IA-2 Ab radioimmunoassay (RIA), IA–2 autoantibodies in patients’ sera, calibrators and controls are allowed to interact with 125I labelled IA-2. After an overnight incubation, solid phase protein A is added to precipitate labelled IA-2 – IA-2 Ab complexes. After a further 1 hour incubation period, assay buffer isadded to each tube and each tube then centrifuged. Any unbound 125I-IA-2 is removed from the tubes by aspiration of the supernatants. The pellets are then counted on a gamma counter and the % binding determined. The level of radioactivity in the pellet is proportional to the amount of IA-2 antibody in the sample. The measuring range is 1 – 50 u/mL.
