链霉蛋白酶消化酶的配制
贮存液浓度20mg／m1，-20℃保存。*佳反应缓冲液为0．01mol／LTris—HCl(pH7．8)，0.01mol／LEDTA和0．5％SDS。常用工作浓度lmg／m1，工作温度37℃。
消化酶的选择
在选择用何种酶消化时，应针对要显示的抗原成分不同而选用不同的酶。一般胃蛋白酶和菠萝蛋白酶消化主要用于细胞间质抗原的检测，例如纤连蛋白(fibronectin)、层黏连蛋白(1aminin，LN)和各型胶原的显示；其余酶消化均可用于细胞内抗原的检测。在配制蛋白酶时，应考虑到酶的*佳pH。例如胰蛋白酶的*佳pH为7．6，要在钙离子的激活下于37℃才能达到*佳效率；而胃蛋白酶则在酸性pH值下才能达到*佳效率。另外，经酶消化后的切片应充分洗涤，以去除残留的消化酶，否则会对抗体或组织进行缓慢消化，影响抗原抗体的结合，破坏组织结构。阳性和阴性对照片应同时消化，避免人为差异。
